O Padre Jesuíta Gabriel (Jeremy Irons) vai para a terra dos Guaranis, na América do Sul, com o propósito de converter os nativos ao Cristianismo.

Rapidamente ele constrói uma missão, juntamente com Rodrigo Mendoza (Robert De Niro), um comerciante de escravos em busca de redenção.

Quando um tratado transfere a terra da Espanha para Portugal, o governo português quer capturar os nativos para o trabalho escravo.

Mendoza e Gabriel protegem a missão, discordando da realização da tarefa.
